The Biosensis Mouse Thrombopoietin/TPO ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody specific for Thrombopoietin/TPO has been precoated onto 96-well plates. Standards and test samples are added to the wells, and then a biotinylated detection polyclonal antibody specific for Thrombopoietin/TPO is added subsequently followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates washed away again with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP driven enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that is changed into yellow by the addition of an acid stop solution. The intensity of this coloured product is directly proportional to the concentration of TPO present in the samples. The purpose of this kit is the in-vitro quantitative determination of mouse TPO in samples such as serum, EDTA or heparin treated plasma, tissue lysates and cell culture supernatants. This kit has been configured for research use only and is not to be used in diagnostic or clinical procedures.
