- Crystallins are separated into two classes:ntaxon-specific, or enzyme, and ubiquitous. The latter classnconstitutes the major proteins of vertebrate eye lens and maintainsnthe transparency and refractive index of the lens. Since lensncentral fiber cells lose their nuclei during development, thesencrystallins are made and then retained throughout life, making themnextremely stable proteins. Mammalian lens crystallins are dividedninto alpha, beta, and gamma families; beta and gamma crystallinsnare also considered as a superfamily. Alpha and beta families arenfurther divided into acidic and basic groups. Seven protein regionsnexist in crystallins: four homologous motifs, a connecting peptide,nand N- and C-terminal extensions. Alpha crystallins are composed ofntwo gene products: alpha-A and alpha-B, for acidic and basic,nrespectively. Alpha crystallins can be induced by heat shock andnare members of the small heat shock protein (sHSP also known as thenHSP20) family. They act as molecular chaperones although they donnot renature proteins and release them in the fashion of a truenchaperone; instead they hold them in large soluble aggregates.nPost-translational modifications decrease the ability to chaperone.nThese heterogeneous aggregates consist of 30-40 subunits; thenalpha-A and alpha-B subunits have a 3:1 ratio, respectively. Twonadditional functions of alpha crystallins are an autokinasenactivity and participation in the intracellular architecture.nAlpha-A and alpha-B gene products are differentially expressed;nalpha-A is preferentially restricted to the lens and alpha-B isnexpressed widely in many tissues and organs. Elevated expression ofnalpha-B crystallin occurs in many neurological diseases; a missensenmutation cosegregated in a family with a desmin-related myopathy.

- Store at 4˚C for three months and -20˚C, stable for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures.
