- The protein encoded by this gene is a member of thenthree-amino acid loop extension (TALE) superclass of atypicalnhomeodomains. TALE homeobox proteins are highly conservedntranscription regulators. This particular homeodomain binds to anpreviously characterized retinoid X receptor responsive elementnfrom the cellular retinol-binding protein II promoter. In additionnto its role in inhibiting 9-cis-retinoic acid-dependent RXR alphantranscription activation of the retinoic acid responsive element,nthe protein is an active transcriptional co-repressor of SMAD2 andnmay participate in the transmission of nuclear signals duringndevelopment and in the adult. Mutations in this gene are associatednwith holoprosencephaly type 4, which is a structural anomaly of thenbrain. Alternative splicing has been observed at this locus andneight variants, encoding four distinct isoforms, are described.
