- Calreticulin is a multifunctional protein that acts as anmajor Ca(2+)-binding (storage) protein in the lumen of thenendoplasmic reticulum. It is also found in the nucleus, suggestingnthat it may have a role in transcription regulation. Calreticulinnbinds to the synthetic peptide KLGFFKR, which is almost identicalnto an amino acid sequence in the DNA-binding domain of thensuperfamily of nuclear receptors. Calreticulin binds to antibodiesnin certain sera of systemic lupus and Sjogren patients whichncontain anti-Ro/SSA antibodies, it is highly conserved amongnspecies, and it is located in the endoplasmic and sarcoplasmicnreticulum where it may bind calcium. The amino terminus ofncalreticulin interacts with the DNA-binding domain of thenglucocorticoid receptor and prevents the receptor from binding tonits specific glucocorticoid response element. Calreticulin canninhibit the binding of androgen receptor to its hormone-responsivenDNA element and can inhibit androgen receptor and retinoic acidnreceptor transcriptional activities in vivo, as well as retinoicnacid-induced neuronal differentiation. Thus, calreticulin can actnas an important modulator of the regulation of gene transcriptionnby nuclear hormone receptors. Systemic lupus erythematosus isnassociated with increased autoantibody titers against calreticulinnbut calreticulin is not a Ro/SS-A antigen. Earlier papers referrednto calreticulin as an Ro/SS-A antigen but this was later disproven.nIncreased autoantibody titer against human calreticulin is found inninfants with complete congenital heart block of both the IgG andnIgM classes.

- Store at 4˚C for three months and -20˚C, stable for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures.
