- Rat Resistin ELISA is a sandwich enzyme immunoassay for the quantitative measurement of rat resistin in serum.Features:? The total assay time is less than four hours. ? The kit measures total serum (homodimeric) resistin.? Assay format is 96 wells.? Quality controls are recombinant rat resistin-based. No animal sera are used. ? Standard is recombinant protein based.? Components of the kit are provided ready-to-use, concentrated or lyophilized.Test Principle: Standards, Quality Controls and samples are incubated in microplate wells pre-coated with polyclonal anti-rat resistin antibody. After 60 minutes incubation and washing, biotin-labelled polyclonal anti-rat resistin antibody is added and incubated for 60 minutes with captured resistin. After another washing, streptavidin-HRP conjugate is added. After 30 minutes incubation and the last washing step, the remaining conjugate is allowed to react with the substrate solution (TMB). The reaction is stopped by addition of acidic solution and absorbance of the resulting yellow product is measured. The absorbance is proportional to the concentration of resistin. A standard curve is constructed by plotting absorbance values against concentrations of Standards, and concentrations of unknown samples are determined using this standard curve.Kit Components:R1585-04A: Microtiter Plate, 1x96wellsR1585-04B: Master Standard, 1x1vialR1585-04C1: High Control, 1x1vial R1585-04C2: Low Control, 1x1vial R1585-04D: Pab (Biotin), 1x13mlR1585-04E: Streptavidin (HRP), 1x13mlR1585-04F: Dilution Buffer, 1x20mlR1585-04G: Wash Solution (10X), 1x100mlR1585-04H: Substrate Solution (TMB), 1x13mlR1585-04J: Stop Solution, H2SO4, 1x13mlStorage and Stability:Store all components at 4oC. Unopened kit components are stable for 6 months. Upon reconstitution of R1585-04D, R1585-04E and R1585-04F, aliquot and store unused portions at -20°C. The aliquots are stable for 3 months at -20°C. Avoid repeated freeze thaw cycles. Other opened kit components are stable for 3 months at 4oC. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
