- Stem Cell Factor (SCF) is a hematopoietic growth factor that stimulates the proliferation of mast cells. SCF also mediates cell-cell adhesion and acts synergistically with other cytokines. The actions of SCF are exerted by binding to the SCF Receptor (PE-1243-50) which results in receptor dimerization. SCF is a type I membrane glycoprotein with 5 potential N-linked glycosylation sites. Soluble SCF is produced by proteolytic cleavage of the extracellular domain.Source:DNA sequence encoding the signal peptide and the mature Stem Cell Factor (SCF) protein of human SCF was expressed in modified human 293 cells.Biological. Activity:The ED50 of SCF is typically 10-20 ng/ml as measured in a cell proliferation assay using the human growth factor-dependent M-07e cell line.Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution:Western Blot: SCF migrates as a broad band between ~20 and 42kD in SDS-PAGE due to post-translational modifications. Unmodified SCF has a predicted molecular mass of 21.0kD. SCF separates into a number of isoforms with a pI between 3.0 and 7.0 in 2D PAGE due to post-translational modifications. This compares with the unmodified SCF with a predicted pI of 5.76. Optimal dilutions to be determined by the researcher.Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
