測定程序

在測定前，讓試劑在室溫下放置。使用前輕輕混合所有試劑。

1. 將所需數量的包被條放入支架中。

2. 將50 微升的C肽標準品、對照品和患者的血清樣本滴入相應的孔中。

3. 向所有孔中加入100 微升的C肽結合試劑。

4. 蓋上板子，在室溫（20-25°C）下孵育60分鐘。

5. 移除所有孔中的液體。用300 微升的1X洗滌緩沖液沖洗孔三次。在吸水紙毛巾上拍干。

6. 向所有孔中加入100 微升的TMB底物。

7. 在室溫下孵育15分鐘。

8. 向所有孔中加入50 微升的終止液。輕輕搖動板子以混合溶液。

9. 在加入終止液后15分鐘內，在ELISA讀數器上以450納米波長讀取吸光度。

結果計算

標準曲線的構建如下：

1. 檢查每個標準品瓶上的C肽標準值。這個值可能會因批次而異。確保你檢查每個試劑盒上的值。參見附帶的標準品示例。

2. 為了構建標準曲線，在線性坐標紙上，將每個C肽標準點的OD值（垂直軸）與C肽標準濃度（水平軸）繪制出來。通過這些點畫出最佳曲線。

3. 從曲線上讀取對照品和每個未知樣本的濃度（納克/毫升）。記錄每個對照或未知樣本的值。
