艾美捷PNA Bio-Cy3標(biāo)記的Cas9蛋白：

左圖：將不同量的Cas9蛋白與500ngsgRNA孵育。通過(guò)脂質(zhì)體胺將RNP復(fù)合物轉(zhuǎn)染到NIH3T3細(xì)胞中。24小時(shí)后，從每種條件下制備DNA用錯(cuò)配敏感核酸酶法（T7E1）檢測(cè)Cas9蛋白和sgRNA的活性。

右圖：對(duì)于體內(nèi)成像，通過(guò)以下方法將500ng的Cas9蛋白和500ng的sgRNA轉(zhuǎn)染到NIH3T3細(xì)胞lipofectamine。24小時(shí)后，通過(guò)共焦顯微鏡對(duì)細(xì)胞進(jìn)行再成像顯微鏡。

Cy3-Cas9蛋白的活性通過(guò)電穿孔進(jìn)入細(xì)胞，然后進(jìn)行T7E1測(cè)定來(lái)確認(rèn)。熒光通過(guò)共聚焦顯微鏡確認(rèn)。
