SiR-微管蛋白是基于微管結(jié)合藥物多西他賽的。SiR-微管蛋白具有熒光特性、細(xì)胞通透性,并且對(duì)微管具有很高的特異性。SiR-微管蛋白能夠在不需要基因操作或過(guò)表達(dá)的情況下對(duì)內(nèi)源性微管進(jìn)行染色。其遠(yuǎn)紅光發(fā)射最大限度地減少了光毒性以及樣本的自發(fā)熒光。SiR-微管蛋白與綠色熒光蛋白(GFP)和/或 m-櫻桃熒光蛋白兼容。可以使用標(biāo)準(zhǔn)的 Cy5 濾光片組進(jìn)行成像。SiR-微管蛋白可用于活細(xì)胞和組織的寬場(chǎng)、共聚焦、SIM 或 STED 成像。探針數(shù)量允許進(jìn)行 50 - 200 次染色實(shí)驗(yàn)。
