試劑盒原理ORAC分析法中的自由基主要來源于偶氮化合物2,2'-偶氮-雙-(2-瞇基丙烷)氯化二氫(2,2'-azobis(2-amidinopropane)dihydrochloride,AAPH)熱分解產(chǎn)生的過氧化氫自由基,也可以是芬頓(Fenton)反應(yīng)過程中產(chǎn)生的羥自由基,以熒光素鈉(Sodium Flourescein,F(xiàn)L)為熒光探針,觀察自由基與熒光探針作用后,探針熒光強(qiáng)度的衰退過程,以水溶性維生素E類似物(Trolox)作為抗氧化標(biāo)準(zhǔn)物質(zhì),檢測(cè)體系中各種抗氧化劑延緩探針熒光強(qiáng)度衰退的能力,以此評(píng)價(jià)抗氧化劑的抗氧化能力。

1.是否可以測(cè)試?yán)鋬霰4娴臉悠罚?/p>

RE:通常適用于在 -80℃ 下保存兩個(gè)月以內(nèi)樣品,但我們建議盡可能測(cè)試新鮮樣品以獲得最佳結(jié)果

 

2.ORAC、HORAC和TAC檢測(cè)之間的區(qū)別,如何選擇?

RE:所有三種測(cè)定都測(cè)量抗氧化能力,但略有不同。ORAC檢測(cè)分析氧自由基的抗氧化能力;HORAC檢測(cè)分析羥基自由基的抗氧化能力;TAC基于單電子轉(zhuǎn)移(SET)機(jī)制和銅離子,檢測(cè)總體抗氧化能力。總體而言,研究人員更偏好選擇ORAC分析而不是HORAC,因?yàn)檫^氧自由基似乎比羥基自由基更相關(guān)。
