小巧、計量精確且可重復(fù)的標(biāo)記
我們采用位點特異性偶聯(lián)化學(xué),確保每個 sdAb 上的熒光染料數(shù)量均一、位置確定。化學(xué)偶聯(lián)完成后,過量的游離染料會被徹底去除。這些生產(chǎn)關(guān)鍵步驟將背景降至最低,并賦予產(chǎn)品最高的批次重現(xiàn)性。配合 sdAb 本身的高親和力單價結(jié)合模式,F(xiàn)luoTags 在所有熒光檢測中均可提供高度線性且可靠的信號。此外,得益于 sdAb 的小尺寸,F(xiàn)luoTags 試劑相比傳統(tǒng)抗體具有更快、更深的組織穿透能力,既能進(jìn)入擁擠的細(xì)胞微環(huán)境,也能在 3D 組織切片及整體樣本中實現(xiàn)快速而均勻的滲透。

FluoTags-Q

所有 FluoTags-Q 試劑均為嚴(yán)格 1:1 化學(xué)計量比的偶聯(lián)物:每個熒光染料精確對應(yīng)一個單域抗體,且熒光染料位于距抗原結(jié)合面 <1.5 nm 的位置。

FluoTags-X2
一個靶標(biāo) – 一個單域抗體 – 兩個熒光染料(1:1:2)
FluoTags-X2 試劑由單個單域抗體攜帶兩個熒光染料構(gòu)成,亮度可達(dá)對應(yīng) FluoTags-Q 產(chǎn)品的 2 倍(“X2”)。熒光染料距抗原結(jié)合面的最大位移約 3 nm。其分子尺寸僅略大于 FluoTags-Q,因此組織穿透性能同樣優(yōu)異。

FluoTags-X4
一個靶標(biāo) – 兩個單域抗體 – 四個熒光染料(1:2:4)
每份 FluoTags-X4 試劑由兩條不同序列的單域抗體(FluoTags-X2 與 FluoTags-X2-B)混合而成,二者可獨立結(jié)合同一靶蛋白。每條抗體各帶兩個熒光染料,總計可將 4 個熒光染料導(dǎo)向單個靶蛋白,亮度可達(dá) FluoTags-Q 的 4 倍(“X4”)。熒光染料距各自抗原結(jié)合面的最大位移仍約 3 nm。分子尺寸與 FluoTags-X2 相同,保持同等優(yōu)良的組織穿透能力。
