Ub-醛是通過將天然泛素中的第76位甘氨酸替換為2-氨基乙醛而合成的。在去泛素化酶(DUBs,例如UCH和大多數(shù)USPs)的活性位點(diǎn)中,Ub-醛的結(jié)合使得活性半胱氨酸的巰基與反應(yīng)性醛基相鄰。巰基對(duì)醛基的親核攻擊會(huì)形成一個(gè)穩(wěn)定的泛素與去泛素化酶之間的加合物。因此,Ub-醛是一種強(qiáng)效的去泛素化酶自殺性抑制劑[1-5]。
