1994年人們首次發(fā)現(xiàn)了瘦素（leptin,LEP），瘦素是一個(gè)由脂肪組織分泌的蛋白類激素，而指導(dǎo)其合成的基因被稱為肥胖基因（obesegene，ob基因）。瘦素在血液中是分子質(zhì)量為16kDa的具有親水性的單體，它是167個(gè)氨基酸在入血時(shí)自行去除掉N端21個(gè)氨基酸信號(hào)肽后所形成。小鼠與人的瘦素受體有將近80%的同源性。經(jīng)研究短型LRa和長(zhǎng)型LRb是發(fā)揮主要作用的兩種受體。

瘦素和瘦素受體與肥胖的發(fā)生有著密切關(guān)系。瘦素受體在下丘腦中廣泛分布,瘦素可以幫助建立、調(diào)節(jié)下丘腦中多條調(diào)控代謝的神經(jīng)通路,影響中樞神經(jīng)系統(tǒng)對(duì)糖脂代謝平衡、能量代謝平衡的調(diào)控。
