纖連蛋白從牛血漿中純化。蛋白質(zhì)純度通過在4-20%聚丙烯酰胺凝膠上掃描考馬斯藍(lán)染色蛋白質(zhì)的密度測定來確定。HiLyte Fluor 488標(biāo)記的纖連蛋白純度為>80%（圖1）。

該蛋白被修飾以含有隨機(jī)表面賴氨酸共價(jià)連接的HiLyte Fluor 488（2）。熒光染料的活化酯用于標(biāo)記蛋白質(zhì)。標(biāo)記化學(xué)計(jì)量是通過蛋白質(zhì)和染料濃度的光譜測量來確定的。最終標(biāo)記化學(xué)計(jì)量是每個(gè)蛋白質(zhì)分子 1-3 種染料（圖 2）。

HiLyte Fluor 488標(biāo)記的纖連蛋白可以使用350-450nm激發(fā)和500-550nm發(fā)射的濾光片組進(jìn)行檢測。纖連蛋白在SDS-PAGE上作為單個(gè)亞基運(yùn)行，表觀分子量為230 kDa。FNR02以橙色凍干粉的形式提供。每瓶FNR02含有20μg蛋白質(zhì)。
