- RAGE, the Receptor for Advanced Glycation Endproducts, is a 35kD transmembrane receptor of the immunoglobulin super family. It is also known as “AGER”. AGER gene is mapped to chromosome 6p21.3 by mapping by contiguous cosmids and YAC clones and by fluorescence in situ hybridization. The expression of RAGE is particularly increased in neurons close to deposits of amyloid beta peptide and to neurofibrillary tangles. RAGE has been linked to several chronic diseases, which are thought to result from vascular damage. The pathogenesis is hypothesized to include ligand binding upon which RAGE signals activation of the nuclear factor kappa B (NF-κB).Intended Use:RAGE BioAssay ELISA Kit is a sandwich ELISA for quantitative detection of rat RAGE in cell culture supernates, serum and plasma (heparin, EDTA).Range: 78-5000pg/mlSensitivity: <10pg/mlSpecificity: Natural and recombinant rat RAGECrossreactivity: No detectable crossreactivity with other relevant proteinsKit Components:144021A: Microtiter Strips, 1x96 wells*144021B: RAGE Standard, 2x1 vial144021C: Anti-rat RAGE (Biotin), 1x130ul143696: Avidin-Biotin-Peroxidase Complex (ABC), 1x130ul (1:100 dilution)143697: Sample Diluent Buffer, 1x30ml143698: Antibody Diluent Buffer, 1x12ml143699: ABC Diluent Buffer, 1x12ml143700: TMB Color Developing Agent, 1x10ml143701: TMB Stop Solution, 2N H2SO4, 1x10mlStorage and Stability:Store *14021B powder at 4°C. Once reconstituted store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Add samples and standards and incubate the plate at 37°C for 90 minutes. Do not wash. 2. Add biotinylated antibodies and incubate the plate at 37°C for 60 minutes. Wash plate 3 times with 0.01M TBS or PBS.3. Add ABC working solution and incubate the plate at 37°C for 30 minutes. Wash plate 5 times with 0.01M TBS or PBS.4. Add TMB color developing agent and incubate the plate at 37°C in dark for 15-20 minutes.5. Add TMB stop solution and read.
