產(chǎn)品描述: The BioAssay?ELISA Kit is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of Vascular Cell Adhesion Molecule 1 (VCAM-1) (Rabbit) in serum, plasma, cell culture supernatants, cell lysates, body fluid and tissue homogenates.Detection Range:5.0-100ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity:This assay has high sensitivity and excellent specificity for detection of Vascular Cell Adhesion Molecule 1 (VCAM-1) (Rabbit). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between the Target Protein and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Vascular Cell Adhesion Molecule 1 (VCAM-1) BioAssay ELISA Kit (Rabbit) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to Vascular Cell Adhesion Molecule 1 (VCAM-1) (Rabbit). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since?VCAM-1?from samples and?VCAM-1-HRP conjugate compete for the anti-VCAM-1?antibody binding site. Since the number of sites is limited, as more sites are occupied by?VCAM-1?from the sample, fewer sites are left to bind the?VCAM-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.
