產(chǎn)品描述: Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades type IV and V collagens. Studies in rhesus monkeys suggest that the enzyme is involved in IL-8-induced mobilization of hematopoietic progenitor cells from bone marrow, and murine studies suggest a role in tumor-associated tissue remodeling.Sample Type:Serum, plasma, tissue homogenates, cell culture supernates and urineIntended Use:For the quantitative determination of human matrix metalloproteinase 9/gelatinase B (MMP-9) concentrations in serum, plasma, tissue homogenates, cell culture supernates and urine. Sensitivity:0.284ng/mlRange:0.312-20ng/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of human MMP-9. No significant cross-reactivity or interference between human MMP-9 and analogues was observed.Test Principle:This BioAssay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for MMP-9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-9 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-9 bound in the initial step. The color development is stopped and the intensity of the color is measured.
