3′-(O-Propargyl)-GTP can be incorporated in RNA at the 3’end using poly(A) Polymerase. But for adding an alkyne to the 3′-end 3′-(O-Propargyl)-ATP (BCT-31) is superior. We developed a different approach in which 3′-(O-Propargyl)-GTP is added to mRNA to the 5’end with an capping enzyme and afterwards methylated. As a result a cap-like structure is created which provides all the advantages of a cap structure together with the possibility to perform click reactions at the incorporated alkyne.
