背景資料: Complete service includes immunizing the HCP preparation after fractionation into high molecular weight (HMW) and low molecular weight (LMW) fractions by ultra filtration (provided by the client). Typically a membrane with a nominal molecular weight cutoff of 30,000 daltons is used to separate proteins less than 30kDa from those greater than 30 kDa. Separately HMW and LMW proteins are immunized into New Zealand White female rabbits. Six rabbits are immunized per immunogen. Project includes: 2 X 6 RABBIT hosts, 90 days daily maintenance, 12 Pre-bleeds (up to 5 mL each), 24 Test bleeds (up to 5 mL each), 20 production bleeds (4 production bleeds from the 5 best responding RABBITS) of up to 25 mL each or up to 500 mL). After a series of immunizations and test bleeds, reactivity against HCPs is assessed by western blot. Only host animals showing reactivity to a broad range of proteins by size are selected for production. Terminal bleeds (1 each from the best responding 5 RABBITS of 100 mL each or up to 500 mL), western blotting of all test bleeds, production bleeds and terminal bleeds and against one (1) HCP lysate provided by client. After selection, host animal production bleeds are pooled and then purified by a method equivalent to Protein A chromatography (7S fractionation or IgY purification from eggs) up to 1.0 L of qualified antiserum. Antibodies to HMW HCP proteins and antibodies to LMW HCP man be combined after purification and validation at the client's request. Vialing and lyophilization of up to 100 glass vials. Final qualification of final product by western blotting against provided HCP lysates. Summary report (1 page) with digital copy of finished product western blot and SDS-PAGE.
