背景資料: Complete service includes immunizing two different host species for anti-HCP antibody production to control for host species variability. Client selects two host animals: rabbit, chicken or sheep. Rabbits are valued as host animals due to the volume of historical data. Chicken/avian host show a varied pattern of immune response to mammalian HCP preparations especially CHO cell derived. Sheep may show greater immune sensitivity and yield larger volumes of antisera. HCP preparation is immunized in the two selected hosts. Project includes: 10 RABBIT or CHICKEN hosts or 3 SHEEP hosts, 90 days daily maintenance EACH, 10 Pre-bleeds EACH (up to 5 mL each), 20 Test bleeds EACH (up to 5 mL each), 8 to 20 production bleeds EACH (4 production bleeds from the 5 best responding RABBITS or CHICKENS or 4 production bleeds from the 2 best responding SHEEP) of up to 25 mL each (65 mL for SHEEP) or up to 500 mL). Only host animals showing reactivity to a broad range of proteins by size are selected for production. Terminal bleeds (1 each from the best responding 5 RABBITS or CHICKENS or 2 SHEEP of 100 mL each or up to 500 mL), western blotting of all test bleeds, production bleeds and terminal bleeds and against one (1) HCP lysate provided by client. After selection, host animal production bleeds are pooled and then purified by a method equivalent to Protein A chromatography (7S fractionation or IgY purification from eggs) up to 1.0 L of qualified antiserum. Vialing and lyophilization of up to 100 glass vials. Final qualification of final product by western blotting against provided HCP lysates. Summary report (1 page) with digital copy of finished product western blot and SDS-PAGE. Package charge for shipping of finished product is included.
