CD5, also known as T1 or LEU1, is a cluster of differentiation found on a subset of IgM-secreting B cells called B-1 cells, and also on T cells. It is mapped to 11q12.2. Although CD5 can be found on both T cells and B cells, T cells express higher levels of CD5 than B cells. CD5 was used as a T-cell marker until monoclonal antibodies against CD3 were developed. CD5 is upregulated on T cells upon strong activation. In the thymus, there is a correlation with CD5 expression and strength of the interaction of the T cell towards self-peptides. Nevertheless, CD5 serves to mitigate activating signals from the BCR so that the B-1 cells can only be activated by very strong stimuli(such as bacterial proteins) and not by normal tissue proteins.

Useful in Sandwich ELISA for Quantitative Detection of Antigen. Aliquot 0.1ml per well of the 6000pg/ml, 3000pg/ml, 1500pg/ml, 750pg/ml, 419pg/ml, 187.5pg/ml, 93.7pg/m human CD5 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of human cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. It is recommended that each human CD5 standard solution and each sample be measured in duplicate.
Store vials at 4°C prior to opening. Centrifuge product if not completely clear after standing at room temperature. This product is stable for 6 months at 4°C as an undiluted liquid. Dilute only prior to immediate use. For extended storage freeze at -20°C or below for 12 months. Avoid cycles of freezing and thawing.
